Downregulation of Myocardial / 3 - Adrenergic Receptors Receptor Subtype Selectivity Kathryn

چکیده
T he desensitization (loss of response) and downregulation (loss of receptors) of p-adrenergic receptors (,BAR) have been observed in several cardiac conditions, including certain forms of heart failure and hypertension. The inability of the failing heart to respond to inotropic agents such as catecholamines represents a major clinical problem. The downregulation of 131AR associated with certain forms of heart failure1 has been considered a consequence of the chronic high plasma catecholamine levels associated with this disorder. The purpose of this review is to examine agonist-induced downregulation of myocardial ,6AR in light of studies suggesting that f31AR are more resistant to downregulation than are /2AR. Potential mechanisms for differential downregulation of flAR subtypes will also be discussed, including the possibility that f82AR on cardiac myocytes are downregulated via coated pits but that f31AR are associated with plasmalemmal vesicles known as caveolae.

متن کاملMechanisms for myocardial beta-adrenergic receptor desensitization in heart failure.
C ongestive heart failure is associated with pronounced alterations in adrenergic nervous system function. Levels of myocardial norepinephrine are decreased, as first reported in 1963,1 a phenomenon later found to be associated with increased sympathetic nervous system activation and high levels of circulating plasma catecholamines2 in combination with abnormalities in neural reuptake of norepi...
متن کاملSelectivity of dobutamine for adrenergic receptor subtypes: in vitro analysis by radioligand binding.
The cardiovascular responses elicited by dobutamine are distinctly different from those produced by other adrenergic or dopaminergic agonists. To test the hypothesis that dobutamine could have differential affinities for adrenergic receptor subtypes, and that such subtype selectivity could be related to its relatively unique pharmacologic properties, we assessed the ability of dobutamine to dis...
متن کاملDesensitization of myocardial beta-adrenergic receptors during cardiopulmonary bypass. Evidence for early uncoupling and late downregulation.
BACKGROUND Cardiopulmonary bypass (CPB), a process routinely used during cardiac surgery, is a potent stimulant to the release of endogenous catecholamines. Hence, we tested the hypothesis that CPB results in myocardial beta-adrenergic receptor (beta AR) desensitization. METHODS AND RESULTS We obtained canine transmyocardial left ventricular biopsies before, during (155 minutes), and after CP...
